Biophysical approach to evaluate DNA transfer by PLGA nanoparticles

A survey of nanoparticles used for drug delivery is provided, focusing on poly(lactic-co-glycolic acid) (PLGA). The interaction between DNA and PLGA nanoparticles has been investigated. A complex structure comprising the nanoparticles and DNA chains has been unambiguously demonstrated, using a new approach involving spectrophotometric and ultracentrifuge methods. It is shown that DNA chains bind only those nanoparticles whose surface has positive charge; DNA molecules will not form a complex with negatively charged nanoparticles. DNA complexes with spherical chitosan-coated PLGA particles by winding around them. The optimal DNA: nanoparticle ratio was found to be 1:7 by weight. This enables one nanoparticle to transfer about 3.5 × 105 base pairs of DNA to the cell cytoplasm.
